چکیده

During the past 2 years microbiological procedures’ have been reported for the determination in proteins of thirteen of the twenty-one amino acids which occur in protein materials. Seven amino acids (isoleucine, glutamic acid, methionine, leucine, phenylalanine, tryptophane, and valine) have been determined with Lactobacillus arabinosus 17-5, three (arginine, tyrosine, and valine) with Lactobacillus casei, two (histidine and lysine) with Leuconostoc mesenteroides P-60, two (aspartic acid and serine) with Lactobacillus delbriickii (4), and one (tryptophane) with Eberthella typhosa T-63 (5). Comparable methods (unpublished) have been developed in the authors’ laboratory for the determination of cystine, glycine, and threonine. It is possible, therefore, to determine microbiologically all of the accepted amino acids except alanine, proline, hydroxyproline, 3,5diiodotyrosine, and thyroxine. It has been shown, previously, that phenylalanine2 is essential for the growth of Clostridium sporogenes (7), Corynebacterium diphtheriae (8), Streptococcus hemolyticus (9)) Lactobacillus mannitopoeus (lo), Lactobacillus buchneri (lo), Lactobacillus lycopersici (lo), Lactobacillus delbriickii (II), Lactobacillus arabinosus 17-5 (12-16), Lactobacillus casei (3,17), and Leuconostoc mesenteroides P-60 (18). Although Hegsted (15) has assayed phenylalanine with Lactobacillus arabinosus 17-5, the present studies were undertaken

Media for Leuconostoc mesenteroides P-60 and Leuconostoc citrovorum 8081.

In the routine use of typical media for the microbiological determination of the sixteen amino acids standard curves have accumulated which reveal the amounts of each amino acid required for the growth of the assay organism when all other amino acids are present in excess. Thus it was observed that the relative amounts of the various amino acids needed by Leuconostoc mesenteroides P-60 did not ...

Identification of lactic acid bacteria isolated from traditional drinking yoghurt in tribes of Fars province

Morphological, cultural, physiological and biochemical characteristics were employed to identify lactic acid bacteria (LAB), isolated from drinking yoghurt in different areas in Fars province, Iran. From 18 drinking yoghurt samples a total of 673 LAB positives were determined, in which 117 (17.38%) and 556 (82.62%) were identified as lactic acid cocci and lactic acid bacilli, respectively. Addi...

Microbiological determination of cystine, systeine, and glutathione in plasma.

In previous reports from this laboratory, a method for the assay of Dand L-methionine in biologic fluids with Leuconostoc mesenteroides P-60 and Lactobacillus fermenti 36 has been described (1, 2). The present communication describes a method for the determination of cysteine plus cystine in plasma. In a report by other workers, using the medium of Dunn et al. (3), the organisms Lactobacillus b...

Microbiological determination of methionine in proteins and foodstuffs.

Kuiken and coworkers (1) reported that methionine is not essential for the growth of Lactobacillue arabinosus 17-5. Later, Hegsted (2), working with the same organism, found that growth failed when methionine was not present in the medium. He called att, ention to the apparent discrepancy in the results obtained in the two laboratories and stated that it was probably due to the use of different ...

Some interrelationships of amino acids in the nutrition of Leuconostoc mesenteroides.

In 1939 Gladstone clearly demonstrated the phenomenon of amino acid antagonism and showed its relationship to the nutrition of the pathogen Bacillus anthracis. Among the interrelationships involving specific amino acids was the ability of threonine to remove the toxic effects of serine and vice versa. Snell and Guirard (1943) noted the inhibition of growth of Streptococcus lactis by glycine, se...
